RESUMO

An acid fraction derived from the evergreen plant of Mastic gum(Pistacia lentiscus L., family Anacardiacae, genus Pistacis L.) has been found to have bactericidal activity against 11 strains of Helicobacter pylori. However its healing potential is significantly reduced by poor solubility and low biological penetration. Therefore, the current problem is to develop a system that provides increased penetration of the drug into the stomach through the epithelial barrier. Foams are especially interesting in this regard. Foams are light systems, they do not swell, on the contrary, they grow in volume, completely covering the mucous membrane. The foams do not require taste correction and are designed to deliver a therapeutic substance from the skin and mucous membranes and provide an effective treatment. The aim of the research was the determination of formulation and development of technology of innovative medicinal form from Mastic gum. Based on biopharmaceutical studies, the formulation of capsules containing a foaming powder composition was determined: Mastic gum 250.0; Sodium lauryl sulfate (SLS) - 9.0; Lecithin - 12.6; Xanthan 4.5; Gelatin 4.5; Isomalt 90.0; Dry egg whites - 41.0; Sodium bicarbonate - 21.0; Citric acid - 6.0; Metocell M102 - 10.0; Talc - 1,4. The technology of preparation of Mastic gum foaming powder containing capsules has been developed.Foaming capsules containing Mastic gum, in artificial gastric juice, with foam-forming ability and foam stability meet the standard requirements for foam systems.

RESUMO

The content of bioactive compounds of perennial, fragrant species of tuberose or Polianthes tuberosa L. representing the genus Polianthes L., asparagus (Asparagaceae Juss) family, highly decorative and widely accepted in the perfumery industry and extremely sensitive to environmental conditions, native to Mexico, was studied based on of raw materials of the flowers of the said plant introduced to five different locations of the BBG with various exposition and soil content by green technologies. Based on the analysis of the soil obtained from all five locations, we can conclude, that the growth and development of the plant along with the quality and amount of received raw materials significantly depend on the rate of soil acidity, humus, and the content of basic feeders. Good results were revealed in the conditions of the experimental plot of fragrant plants with well enough warmth, sunlight, and humidity. Applying 100% natural bio preparation GeoHumate had a positive influence on the plants on the experimental plot in terms of the growth and development of the plant and accumulation of bioactive compounds. After doing GC-MS researches of tuberose, Polianthes tuberosa L., flowers introduced by green technologies in five different locations of the BBG with various exposition and soil content, there were identified important compounds valuable for cosmetics, perfumery, medicine, and of different fields.

RESUMO

In the present article is considered antihypertensive and cardioprotective action of epoxieicosatrienoic acids (EETs) analogues and soluble epoxide hydrolase (sEH) inhibitors. Being epoxygenase products of arachidonic acid metabolism EETs provide a wide spectrum of biological activity in different organs implicated in the regulation of arterial pressure, including vascular endothelium (VE), heart and kidney. EETs are acting as autocrine or paracrine agents producing vasodilation, reduction in salt and fluid retention. EETs exert vasodilatory properties by activation of the smooth muscle large conductance Ca2+ -activated K+ channels in different vascular beds. In experimental and clinical studies EETs provide beneficial influence in hypertensive states alleviating vascular endothelium function associated with reduction of inflammation and increased Na+ excretion, prevented cardiovascular and renal complications accompanied arterial hypertension (AH). In animals studies using Dahl-salt-sensitive (Dss) hypertensive rats EETs analogues displayed renoprotective effect, reducing mRNA expression of tumor growth factor-"B" as well as concomitant oxidative stress and fibrotic changes in the heart and kidneys. In cultered endothelial cells also was demonstrated anti-inflammatory action of EETs when they significantly decreased TNF-α-induced high level of monocyte chemoattractant protein-1, which was reversed by EETs antagonist. sEH is another new target for therapeutic intervention in cardiovascular diseases. In several experimental models of AH sEH inhibitors (sEHI) significantly reduce AP which was associated with their anti-inflammatory and renoprotective action. sEHI showed cardiovascular effect related to CYP2J2overexpression during myocardial ischemia-reperfusion injury. It should be noted that EETs demonstrate binding ability to PPAR receptor "Y" stimulating it's transcription process in endothelial cells facilitating by sEHI. It is suggested that EETs and sEH are involved in the regulation of the cardiovascular function playing significant role in vascular homeostasis.

RESUMO

The aim of the study was to develop and characterize the nanocomposite in-situ hydrogel as local drug delivery system of cytotoxic agent. In-situ hydrogel consisting of 15% thermosensitive (Poloxamer 407) and 1% mucoadhesive (sodium alginate) polymers was selected as the optimal formulation by the conducted studies. The influence of nanoparticle concentration on gelation time and temperature has been experimentally established. As a result, the optimum concentration of nanoparticles (5%) is selected, which does not alter the gel forming process. The resulting nanocomposite hydrogel was characterized through Fourier transform infrared spectroscopy (FT-IR), scanning electron microscopy (SEM), rotational viscometer (LVDV-1T). FT-IR spectra confirmed the PLGA nanoparticles presence within the hydrogel matrix through the absorption peak located at 1750 cm-1. SEM images allowed observing the nanoparticles to be homogenously dispersed. The release pattern of the active substance from the nanocomposite hydrogel is following: at 72 h, 64% and 78% of the active substance were released into the phosphate buffer and cell culture area, respectively. Irritation test on hen's egg model revealed that formulated nanocomposite hydrogel did not show damage of vascular system.

RESUMO

In the world today, the demand for the preparations and cosmetics, made of peloids is increasing significantly, which is explained by the increased interest of the community to the ecologically clean raw materials of natural origin, it often replaces expensive chemical preparations, which are often accompanied by some contraindications. The increased interest in peloids in the world put on the agenda the question of rational use of acting mud mines as well as the problem of cosmetic and medicinal preparations, developed on their basis. It is noteworthy that the development of ready-made, soft drug forms from the Adjara region sulfide silt peloids is one of the most topical and important issues, as at present there is not available the form of ready-made drugs containing the sulfide silt peloids of this region. Therefore, the aim of the research was to develop the formulation and technology of the hydrogel, containing the Adjara region sulfide silt peloid. The sulfide silt peloid of the Ardagani lake have been used for preparing the hydrogel compositions, as it is distinguished by its advantageous physical-chemical properties and content of biologically active ingredients compared with other studied peloids. The main physical-chemical and technological characteristics of hydrogels compositions have been studied using the following methods: uniformity, pH, the colloidal stability, thermal stability , osmotic activity - by dialysis method using semiconductor membrane (Kruvichinski method), release of active ingredient was studied by using Franz diffusion cells, viscosity was determined by Viscometer RVDV-1 T (Dongguan Zhongli Instrument Technology Co., China), the structure of hydrogel composition - by microscopic (ROW Rathenow, Germany) examination. Based on the conducted studies have been established, that the hydrogel composition N6 is distinguished by its best properties compared to other hydrogel compositions, in which the carboxymethylcellulose was used as a base. The conducted studies became the basis for the development of the formula and technological scheme of production for the hydrogel of Ardagani lake sulfide silt peloid of Adjara region. The results of determining the main characteristics of the given gel provide the desired quality and efficiency of the product.

RESUMO

Medicinal muds, so called Peloids in the last decade attract attention in many countries as effective means for the treatment- preventive, and perfumery-cosmetic purposes. It has been established that the possibilities of this formation of nature are not limited to the many-year experience of the mankind. In terms of further improvement of the human social sphere, nowadays peloids are referred to as XXI century material. The aim of the research was to study the physical-chemical characteristics, such as: humidity, pH, ash content, bulk (volumetric) density and organoleptic properties of sulphide silt peloids of some Adjara region lakes, as well as to determine the content of mineral and organic compounds (humic and organic acids). The objects of the study were the sulphide silt peloids of some Adjara region lakes: Mtsvane, Gasaliani, Shratiani, Nurigeli, Ardagani and Niphi. The main good quality indicators of the sulphide silt peloids have been determined according to the methods described in the State Pharmacopoeia XI. The mineral and organic compounds in the research objects have been determined using the modern instrumental methods of analysis described in the literature. Based on the performed studies, the main physical-chemical characteristics of sulphide silt peloids of some Adjara region lakes (Mtsvane, Gasaliani, Shratiani, Nurigeli, Ardagani and Niphi) have been studied and their mineral contents (Ca-Na Feldspar, K Feldspar, Ca- Montmorillonite, Hematite, Quartz, Magnetite, Chlorite) determined. By the spectrophotometric and gravimetric methods of analysis there is established, that the sulphide silt peloids of the lakes Ardagani (1,22%), Niphi (1,72%) and Gasaliani (1,33%) are distinguished by relatively higher content of humic acids. By the Gas Chromatography-Mass Spectrometry - method of analysis, in the study objects there is established the content of complex ethers of the following organic acids: Stearic, Palmitic, Caprylic, Myristic, Valeric, Lauric, Oxalic, Phthalic and Fumaric acids.

RESUMO

Application of phytofilms based on biosolublepolymers is considered as a prospectivemethod for burn treatment . Herbal remedies contain biologically active substances, that are relatively less toxic, do not cause skin irritation or allergic reactions and, importantly, affectstrains of the microorganisms and viruses resistant to antibiotics and synthetic drugs. Nowadays, the advantages are given to such burn healing drugs, which along with high specific efficacy, have analgesic, anti-inflammatory and antimicrobial effects, and don't irritate the tissues. The mentioned peculiarities are characteristic for a new herbal phenolic biopolymer poly[3-(3,4-dihydroxyphenyl) glyceric acid](PDGA), isolated from the roots and stems of different comfrey species . The aim of the study was the development of the formulation and technology of biosoluble films for burn treatment on the basis of PDGA. The optimal content of phytofilm for burn healing was selected on the basis of the biopharmaceutical study results. The impact of the film-former on the quality, adhesion and moisture absorption of the phytofilmhas been studied. The optimal degree of the phytofilm moisture, determining its high adhesive properties, was established. The film prepared on the basis of sodium alginate, with 30.4% humidity, demonstrated the greatest adhesion strength. After investigation of the PDGA release it was found, that the hydrophilic bases such as: sodium carboxymethyl-cellulose (69.2%) andsodium alginate (78,65%) appeared to be optimal among the others. At the same time, taking into consideration the disadvantages of sodium carboxymethyl-cellulose (tautening effect on burnt surface, relatively low stability), a film based on sodium alginate has been chosen. The manufacturing technology for obtaining PDGA-containing phytofilm by casting is proposed. Theshelf-lifeofproposedPDGA-containingphytofilmis 2 years.

RESUMO

According to the results, investigated methanolic extracts of endemic plant from Adjarian floristic region of Georgia do not have specific cytotoxicity against the cell lines of breast adenocarcinoma MCF-7 human, which is typical, e.g., for plant pigment curcumin. At the same time, the high specific cytotoxicity of 2 and 3 fractions towards keratinocytes gives reason for considering endemic plant extracts, of Adjarian floristic region of Georgia, as a potential pharmacological means for topical treatment of pathologies caused by increased proliferation of keratinocytes, such as psoriasis.

RESUMO

One of the most actual problems of pharmacy is the development of medication forms for external application with complex effects on (gel, emplastro, aerosol, etc.) skin wounds, burns and inflammatory factors. The centuries-old practice of using phyto-preparations (herbal remedies) proved that they have fewer side effects in comparison with synthetic drugs. Despite the wide application of herbal preparations, in the literature there is a little information about their application in development of wound and burn healing modern dosage forms. Among the medicinal plants with the mentioned pharmacological actions, comfrey (Symphytum L.) should be distinguished. Phenolic polymer poly[3-(3,4-dihydroxyphenyl)glyceric acid] (PDGA) or poly[oxy-1-carboxy-2-(3,4-dihydroxyphenyl)ethylene], amounting approximately 25% of polysaccharides and 1.5-2.5% of dry plant material, were isolated from the roots and stems of Caucasian comfrey species (S. asperum, S. caucasicum). Contrary to polysaccharides this phenolic polymer of Comfrey appeared to have a high immunomodulatory (anticomplement), antioxidative, antilipoperoxidantive, anti-inflammatory and wound-healing efficacy/activities. The aim of the study was development of the composition and technology of PDGA-containing gel. According to the results of complex biopharmaceutical studies PDGA gel optimal composition has been proved. The technological scheme for preparation of PDGA gel has been developed. PDGA gel stability under normal conditions of storage at +40С was studied. The gel has a shelf life (determined expiration date) of 2 year.

RESUMO

Perilla is the genus of herbaceous plants of Lamiaceae family. The essential oil of Perilla is believed to be essential part of the herb. It was claimed to have anti-inflammatory, anti-hyperlipidemia, antioxidative and antimicrobial activities. Apart from pharmaceutical and nutrition purposes, It is an edible plant frequently used as a fresh vegetable. It was also applied to produce perfume and cosmetics. Due to the importance of the essential oil from the P. nanakinensis and the lack of detailed studies of them, our work aimed investigation of contents of essential compositions by GC- TOF/MS method. The materials of the study were the aerial parts of P. nankinesis collected at full flowering stage, Guria Region, Georgia. A qualitative analysis of the individual compounds in P. nankinensis essential oil performed by gas chromatography (GC) coupled with time-of-flight mass spectrometer (TOF/MS) for the identification of the resolved peaks. 28 components, which accounted for 77,7% of the oil, were identified. The main components of this essential oil were 1,3,6,10-Dodecatetraene, 3,7,11-trimethyl-, (Z,E)- (α-Farnesene) (34.3%), Caryophyllene oxide (10.2%), 1,6-Octadien-3-ol, 3,7-dimethyl- (linalool) (10.2%). Essential oil also contained humulene (3.9%),caryophyllene (1.9%), methyl salicilate (1.15%), 2-hexanoylfuran (1.0%), gernacrene (1.1%). Based on the results of our experimental data and literature reviews, Perilla essential oil can represent special interest and can be used as an important natural substance for obtaining medicinal and preventive products with the several biological activities.

RESUMO

Vinca roseae L. (Саtharanthus rosea (L.) G. Don) was introduced at Kobuleti experimental station of medical plants. The object of investigation was the plant material of Vinca roseae L. collected in May, 2005., September, 2006 and October, 2009. Total alkaloids were obtained in accordance with Atta- ur-Rachman method. The variability of the quantitative and qualitative composition of total alkaloids and vincaleikoblastin (VLB) fraction during vegetation was studied. It was established that the maximal content of total alkaloids and VLB fraction of Vinca roseae L. is accumulated in the phase of secondary flowering, hence the collecting of a plant material is recommended to be made during the aforesaid vegetation phase as for this period it is a rather high output of a raw material, alkaloid complex and VLB faction. Alkaloids vinkaleikoblastin, ajmalicine and new epimer tetrahydroalstonine with С3Н-α- orientation were yielded, separated and identified using modern physical-chemical and spectral methods (13С NMR).

RESUMO

Comfrey (Symphytum L.) is used to treat bone fractures, tendon injuries, ulcer lesions of gastrointestinal tract. It promotes wound healing, accelerates exudates resorption in lungs and reduces joints' inflammation. In Georgian folk medicine, herbal remedies from comfrey are used to accelerate regeneration processes. Comfrey contains hepatotoxic and carcinogenic pyrrolizidine alkaloids, besides the main active ingredient is poly [3 - (3,4-dihydroxyphenyl) glyceric acid] (PDPGA). The aim of present work was to develop a technology for the substance - poly [3-(3,4dihydroxyphenyl) glyceric acid] (PDPGA) from comfrey stems, free of toxic pyrrolizidine alkaloids. During the investigation the optimal conditions for extraction and purification have been established: on the first stage pyrrolizidine alkaloids were removed from plant material by supercritical extraction; then the crude polysaccharides' fraction was obtained by water extraction (raw materials/extragent ratio was 1:15 at 90oC, the procedure was carried twice for 60 and 90 minutes). The isolation of the final product - PDPGA from crude polysaccharides' fraction was carried out by ultrafiltration on membrane filters. Based on the results of the investigation the technological scheme for the substance has been developed.
